I also have this problem with Autoruns (version 12.01). I have just installed it on my Windows 7 SP1 machine. I tried unchecking several files and got the message mentioned by byphz above every time. For example, "Skype Click to Call for Internet Explorer". The location given by Autoruns is "c:\program files (x86)\skype\toolbars\internet explorer x64\skypeieplugin.dll", and the file is there. However, attempting to uncheck it produces the error.

Granted, the proper way to disable that extension is through "Manage add-ons" on the IE Tools menu, but if Autoruns is going to display it with a checkbox , then I expect to be able to uncheck it. Using "Run as administrator" has no effect with this.
